Drug survival of anakinra and canakinumab in monogenic autoinflammatory diseases
A recent international multicentre collaboration from the AIDA network has conducted a detailed analysis on drug survival of anakinra and canakinumab in monogenic autoinflammatory diseases.

We enrolled 78 patients for a total of 102 treatment regimens. Persistence rate of interleukin-1 inhibitors at 12, 24, and 48 months of follow-up was 75.8%, 69.7% and 51.1%, respectively. Patients experiencing an adverse event and those carrying low-penetrance mutations showed a significantly lower persistence rate. Anakinra and canakinumab display an excellent long-term effectiveness in terms of drug survival, which in turn is not influenced by the preceding biologic treatments. A special attention should be paid to the safety profile and to the genetic diagnosis since patients with low-penetrance mutations are more likely to withdraw from treatment.
